I am not commuting from the Twin Cities (since I am going to move to Rochester), but I have to go back and forth between the two places to take care of new employment stuff at Mayo and the transportation I use is the: Rochester shuttle service (you can search that in google)
This is the only transportation I found that would drop be off directly at Mayo, but it also costs a lot of money

Also, once you start, the commuter bus options are subsidized and I believe you can choose a 10-ride punch pass, monthly, or annual pass option.
